T cell IL-2 and IL-4 release assay

Human CD4+ T-cells are presented with beads coated in CD2, CD3 & CD28, to induce an inflammatory response, detected as an increase in IL-2 & IL-4 secretion. Your compound is tested for the ability to inhibit this inflammatory response, measured as an inhibition of IL-2 & IL-4 release. Cellular proliferation is also assessed, as a measure of general compound cytotoxicity.

T cell IL-2 and IL-4 release assay procedure

The assay utilizes primary human CD4+ T cells seeded at 4000 cells/well in a 384-well microtiter plate and stimulated using CD2, CD3 & CD28-coated antibodies to induce increased secretion of IL-2 and IL-4. The test molecule is added in different concentrations and after 48 hours the IL-2 and IL-4 levels, as well as the cell viability are measured as the final end point.
